Not a solution, but some workarounds to try:

Try opening a command prompt (may need to run as admin) and running copy "C:\ProgramData\Microsoft\Windows\Start Menu\Programs\PowerToys (Preview)\PowerToys (Preview).lnk" "%APPDATA%\Microsoft\Windows\Start Menu\Programs\Startup"

This puts a shortcut to Powertoys into the shell:startup folder. It's how you can manually make programs run at startup.

Alternatively you could run the elevated command schtasks /create /tn "PowerToys" /tr "C:\Program Files\PowerToys\PowerToys.exe" /sc onstart /rl HIGHEST to add it to Task Scheduler's startup tasks.

I am the original author. I can happily say today that Power Toys starts up correctly on both my work and personal computers :)
